The O-specific polysaccharide isolated from the non-capsulated Escherichia coli O125: K-:H19 lipopolysaccharide contains L-fucose, D-mannose, D-galactose, D-glucos and 2-acetamido-2-deoxy-D-galactose in the molar retios 1:1:1:1:1.7. The primary structure of the O-specific polysaccharide was established by compositional analysis, methylation studies, enzymatic hydrolysis and with the 1H and 13C NMR spectroscopy. These studies revealed that the O-antigen is branched and built up of repeating hexasaccharide unit having th following structure: (see struc. in text). This structure is similar to that of recently reported during the course of our investigation for the O-antigenic polysaccharide for a capsulated strain of Escherichia coli O125:K72:H19 of European origin.
