  The lipids extracted with CHCl3/MeOH mixtures from mycelium of the lower filamentous fungus Absidia corymbifera F-295 were found to contain three glycolipids. Based on the IR, H-1 and C-13 NMR spectra, plasma-desorption ionisation (PDI) mass spectra as well as chemical degradation results, the glycolipids were established to be 1-O-β-D-glucopyranosyl-2-N-(2'-D-hydroxyhexadecanoyl)-9-methylsphinga-4(E),8(E)-dienine (glucosyl ceramide) and 2-O-(6'-O-β-D-galactopyranosyl)-β-D-galactopyranosides of 2-D-hydroxy and erythro-2,3-dihydroxy fatty acids C-9, C-11, and C-13. They accounted for about 3.4, 0.8, and 0.4%, respectively, of the total lipids extracted. No lipids identical to the above monohydroxy and dihydroxy fatty acid glycosides have been reported.   Background: Natural products acquire vast and intriguing structural diversity and have been recognized as a tremendously diverse source of new lead compounds. Numerous bioactive secondary metabolites are present in the form of glycosylated molecules in which the sugar parts are normally associated with the interaction along with molecular recognition of the cellular target. Scope and approach: The presence of sugar entities are crucial as well as in some cases necessary, for therapeutic effects. Establishing novel and potent glycosylated secondary metabolites has formed a main goal in the natural product field from fungi and bacteria. These compounds possess a diverse range of sugar units. Key findings and conclusions: Fungi is considered one of the important sources for approved drugs with a diverse range of mode of action. The sugar part in numerous pharmacologically active natural products enhances bioavailability, biological potential, reduce toxicity, and improve stability. The vast majority of glyocosides showed antimicrobial effects, cytotoxic, antiviral and antiinflammatory effects. Notably, numerous fungal glycosides presented in this review illustrate significant antimicrobial effects towards various microorganisms especially against plant pathogens. The antimicrobial effects of these fungal glycosides indicate that these metabolites could be employed as natural preservatives in food in order to abolish or control the growth of pathogenic and spoilage microorganisms.

  Termitomycesphins E and F, novel cerebrosides that are hydroxylated around the middle of the long-chain base (LCB), have been isolated from the edible Chinese mushroom Termitomyces albuminosus (Berk.) Heim. ('Jizong' in Chinese) together with termitomycesphins A-D, and shown to induce neuronal differentiation in rat PC12 cells. Their stereostructures have been determined based on their chemical derivatization and spectroscopic analysis. The major cerebroside obtained from the same mushroom was not hydroxylated around the middle of the LCB and was inactive against PC12 cells, suggesting the importance of the extra hydroxyl group on LCB. The Di- and tetrahydroxylation of this inactive cerebroside resulted in the enhancement of its neuritogenic activity.

  The edible fungus Phellinus baumii Pilat (Hymenochaetaceae) has been used in Korean traditional medicines for strengthening health and prolonging life. An extract of the fruiting bodies of P. baumii was subjected to bioassay-guided fractionation based on its anti-inflammatory effects in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. The resulting fractions were chemically investigated, leading to isolation of three phenolic compounds (1-3), a sesquiterpene (4), two steroids (5-6), a fatty acid (7), and a cerebroside (8). Spectroscopic analyses including 1D and 2D NMR spectroscopy and LC/MS were used to determine their chemical structures. Compounds 2, 4, 5, 7 and 8 were identified in P. baumii for the first time. Since all compounds were isolated from active fractions with anti-inflammatory activity, their ability to inhibit LPS-stimulated nitric oxide (NO) production in RAW264.7 cells were evaluated in vitro. Compounds 1, 2, 3, 5 and 7 inhibited LPS-stimulated NO production, and compounds 1-3 had IC50 values <10 μM. Treatment of LPS-stimulated RAW264.7 cells with compounds 1-3 inhibited phosphorylation of IKKα and IκBα. In addition, treatment of compounds 1-3 reduced LPS-induced increases of nuclear factor-kappa B (NF-κB) p65, iNOS and COX-2 protein expressions. Collectively, compounds 1-3 inhibited NF-κB-dependent inflammation in RAW264.7 cells. Thus, P. baumii is a potential source of natural anti-inflammatory agents, and active compounds 1-3 could be promising lead compounds for the development of novel anti-inflammatory agents.

  Chemical investigation of the methanolic extract from the fruiting bodies of Hericium erinaceus, led to the isolation of fifty-one compounds including thirty-five ergostane-type sterols (1-35), fourteen aromatic compounds (36-49), and two cerebrosides (50 and 51). Their structures were identified based on spectroscopic analyses and by comparison of their spectral data with those reported in literature. This is the first comprehensive low-polarity chemical investigation of H. erinaceus. Thirty-one of the compounds (6-8, 11-35, 39, 41, and 49) were isolated for the first time from the genus Hericium and the family Hericiaceae. The chemotaxonomic relationship between H. erinaceus and other Hericium species was also discussed.

  Pleurotus cornucopiae (Pleurotaceae) is an edible and medicinal mushroom widely distributed in Korea, China, and Japan. The MeOH extract of the fruiting bodies of P. cornucopiae showed renoprotective effects against cisplatin-induced kidney cell damage. Chemical investigation of the MeOH extract led to the isolation and identification of 12 compounds including noransine (1), uridine (2), uracil (3), (3β, 5α, 6β, 22E, 24S) -ergosta-7, 22-diene-3, 5, 6, 9-tetrol (4), (22E,24S)-ergosta-7,22-diene-3β,5α,6β-triol (5), (22E,24R)-ergosta-8(14),22-diene-3β,5α,6β,7α-tetrol (6), cerebroside B (7), (2R) -N- [(1S, 2R, 3E, 7E) -1- [(β-D-glucopyranosyloxy) methyl] -2-hydroxy-8-methyl-3, 7-heptadecadien-1-yl] -2-hydroxy-heptadecanamide (8), cerebroside D (9), nicotinamide (10), 1,2-bis(hydroxymethyl)-4,5-dimethoxybenzene (11), and benzoic acid (12). Among them, compounds 1 and 11 were isolated as naturally occurring products for the first time, though they were reported as synthetic products in previous papers. All of the compounds (except 8 and 11) abrogated cisplatin-induced LLC-PK1 cell damage in a dose-dependent manner. Of special note, compounds 2, 5, 6, and 12 ameliorated cisplatin-induced nephrotoxicity to 80% of the control value at 10μM. The protective effects of compounds 2, 5, 6, and 12 were mediated via the deactivation of JNK-caspase 3 apoptotic cascade. This study is the first to demonstrate that the chemical constituents of P. cornucopiae display renoprotective effects against anticancer drug-induced damage in kidney cells.

  A cerebroside prepared from Schizophyllum commune and shown to have fruiting-inducing activity on the same fungus was studied chemically and biologically. Chemical degradation, which was followed by extensive applications of nuclear magnetic resonance and mass spectrometry to the derivatives, suggested a structure (4E,8E)-N-D-2’-hydroxypalmitoyl-1-O-β-D-glucopyranosyl-9-methyl-4,8-sphingadienine. The degradation products and the derivatives were submitted to a fruiting-inducing assay. The reduced periodate-oxidized cerebroside and the ceramide were as active as the original cerebroside, but the hydrogenated cerebroside and the hydrogenated ceramide were less active and the N-acetyl and N,O-acetyl sphingoids were much less active. The glycoside, the fatty acid, the psychosine, the sphingoid, the acetonide of the N-acetyl sphingoid, and the dinitrophenyl derivative of the hydrogenated sphingoid exhibited no activity. It was concluded that both the methyl branch of the sphingoid and the acid amide linkage in the cerebroside may be the functional structures of the fruiting-inducing substance.

  A new cerebroside, cerebroside E (1) was isolated from the fruiting bodies of Hericium erinaceus (Hericiaceae). The structure of 1 was elucidated by a combination of extensive spectroscopic analyses, including extensive 2D NMR, HR-MS, and chemical reactions. Compound 1 was evaluated for its applicability to medicinal use in several human diseases using cell-based assays. As a result, compound 1 attenuated cisplatin-induced nephrotoxicity in LLC-PK1 cells and exhibited a significant inhibitory effect on angiogenesis in HUVECs. These results collectively reflect the beneficial effects of compound 1 in cancer treatment

  So far, more than 20 species of Ganoderma (Lingzhi) fungi have been studied, including G. lucidum, G. sinense, G. japonicum, G. capense, G. australe, G. tsugae, G. applanatum, G. tropicus, G. boniense, G. duropora, G. resinaceum, G. theaecolum, G. cochlear, G. atrum, G. formosanum, G. boninense, G. colossum, G. concinna, G. amboinense, G. pfeifferi, and G. orbiforme. Over 600 compounds were isolated and identified from the genus Ganoderma. The components in Ganoderma contained triterpenes, meroterpenoids, steroids, alkaloids, nucleosides, nucleobases, and polysaccharides, in which triterpenes were the main compounds, and more than 300 triterpenes obtained from it. With the use of new technology of separation and purification, the other types of compounds, such as meroterpenoids, alkaloids, nucleosides, and polysaccharides were reported in recent years. Herein, the structural classifications and characteristics and separation methods were summarized.

  The detailed chemical analysis of the methanol extract of Meripilus giganteus (Pers.) P. Karst. led to the isolation of two new cerebrosides, mericeramides A (1) and B (2) together with cerebroside B (3), ergosterol (4), 3β-hydroxyergosta-7,22-diene (5), cerevisterol (6), 3β-hydroxyergosta-6,8(14),22-triene (7), 3β-O-glucopyranosyl-5,8-epidioxyergosta-6,22-diene (8) and (11E,13E)-9,10-dihydroxy-11,13-octadecadienoic acid (9). The structures of the compounds were determined on the basis of NMR and MS spectroscopic analysis. Mericeramide A (1) is the first representative of halogenated natural cerebrosides. The isolated fungal metabolites 1-9 were evaluated for their antioxidant activity using the oxygen radical absorbance capacity (ORAC) assay. Compounds 2, 5 and 9 proved to possess considerable antioxidant effects, with 2.50 ± 0.29, 4.94 ± 0.37 and 4.27 ± 0.05 mmol TE/g values, respectively. The result obtained gives a notable addition to the chemical and bioactivity profile of M. giganteus, highlighting the possible contribution of this species to a versatile and balanced diet.
