Herbal medicinal interaction on the purgative action of Chungpesagan-tang, which is one of the traditional Korean medical prescriptions that has been most frequently used for stroke was investigated. Chungpesagan-tang had more potent purgative activity than Rhei Rhizoma alone. When each Chungpesagan-tang composing herbal medicine with Rhei Rhizoma was extracted with water, water extract of Raphani Semen with Rhei Rhizoma had the most potent purgative activity. However, Raphani Semen had not purgative activity. When Rhei Rhizoma with Raphani Semen was extracted, sennoside A content in the water extract was increased. The transformation of sennoside A of Rhei Rhizoma to rheinanthrone by human intestinal microflora was weakly induced by Raphani Semen. These results suggest that the purgative action of Chungpesagan-tang could be controlled by the addition and subtraction of Chungpesagan-tang-composing herbal medicines.

The aim of this study was to clarify whether sennosides were contained in dietary supplements containing senna stem and in the raw materials of senna stem for dietary supplements. The identification and determination of sennosides were performed with thin layer chromatography and high performance liquid chromatography. Moreover, plant contents of herbal teas were classified into leaf, stem and others, and these were identified by comparison with characteristic components of senna using a stereoscopic microscope. The amount of sennosides ranged from about 0.2 mg to 11 mg in a dose of dietary supplements. In three of these 11 samples, senna leaves and midribs were contained. However, in eight of the samples, the plant fragments were not recognized. In five raw materials of senna stem, the amounts of sennosides were from 9.5 mg/g to 14.6 mg/g. These raw materials of senna stem consisted not only of senna stem, but also senna leaves and midribs. We believe that sennosides of dietary supplements are derived from senna leaves and midribs, which contaminate the raw materials of senna stem.

For immunization, sennoside A was conjugated with bovine serum albumin. The hapten number in an antigen conjugate was determined to be five by matrix-assisted laser desorption/ionization TOF mass spectrometry. A hybridoma secreting monoclonal antibody against sennoside A was produced by fusing splenocytes immunized with sennoside A–bovine serum albumin and a hypoxantine–aminopterin–thymidine (HAT)-sensitive mouse myeloma cell line, P3-X63-Ag8-653. Weak cross-reactivities occurred with sennoside B which is a stereochemical isomer, and a monomer of sennoside A, rhein, but no cross-reactivities were observed with other related anthraquinones and phenolics. The full range of the assay extends from 20 to 200 ng/ml of sennoside A. Good correlation of sennoside A concentrations in crude extract of rhubarb between ELISA and HPLC methods was obtained. The contents of sennoside A in various rhubarb roots were assayed by the newly established ELISA indicating a good agreement with the previous reports.

Senna is a well-known drug, used in the Ayurvedic and Allopathic systems of medicine, and is a treatment for constipation. The purgative action of senna and its formulations is due to the presence of sennosides A and B. An HPTLC method has been developed for the determination of individual sennosides (A, B, C, D) without any derivatization in marketed formulations (three tablet formulations, two granule formulations and one liquid formulation) and plant materials (senna leaf and pod). The methanolic solution of a sample was applied on a pre-coated silica gel G60 F254 TLC plate (E. Merck.) and was developed using n-propanol : ethyl acetate : water : glacial acetic acid (3 : 3 : 2 : 0.1 v/v) as the mobile phase. The relative band speeds (Rf values) obtained were 0.35, 0.25, 0.61, 0.46 for sennosides A, B, C and D, respectively. The densitometric response was monitored at 366nm. Calibration curves were found to be linear in the concentration ranges 193-1356, 402-2817, 71-497 and 132-927 ng per spot for sennosides A, B, C, and D, respectively. The correlation coefficients were found to be 0.9978, 0.9987, 0.9939 and 0.9983 respectively for sennosides A, B, C and D. The result obtained with the HPTLC method for total sennoside content was compared with the results using the pharmacopoeial methods (spectrophotometric (British Pharmacopoeia) and spectrofluorimetric (United States Pharmacopeia) using the 'F' test). The results revealed no significant difference in the three different methods for estimation of total sennoside. The proposed HPTLC method was found to be simple, specific, precise, accurate and rapid. It can be used for routine quality control of sennosides or senna-containing formulations for individual sennosides.

The paper reports the development and scale up of new film-coated tablets containing dry herba extract, from the Senna plant, containing sennoside A and B. Film coated tablets containing 10 and 20 mg active ingredient were developed. They had the same disintegration, dissolution and stability behaviours as a reference sugar coated dragee.
